ABSTRACT

Hemodialysis, along with chronic kidney disease (CKD), intensifies the inflammatory process and oxidative stress in patients undergoing treatment. In this context, Vitamin E supplementation can mitigate the deleterious effects resulting from these processes. This is a systematic review whose objective was to evaluate the effect of Vitamin E supplementation on inflammatory biomarkers and oxidative stress in patients with CKD on hemodialysis. This review was prepared according to the methodology for systematic reviews and meta-analyses (PRISMA) and the search was performed in Pubmed, Cochrane Library, Scopus and Web of Science databases. The risk of bias of the included studies was assessed with the Cochrane Risk of Bias Tool. Twelve studies developed between 2006 and 2020 were included in this review. Most of them (n= 11) used vitamin E doses ranging from 400 IU to 888 IU and the supplementation time ranged from 2 weeks to 12 months. Of all the 12 articles included, 25% (n= 3) analyzed supplementation on biomarkers of oxidative stress and 25% (n= 3) addressed these parameters simultaneously. A positive effect was observed in 58.4% of the studies (n= 7). Thus, Vitamin E supplementation can be effective in mitigating the inflammatory process and oxidative stress, however, it is worth noting that the effect depends on the dose and time of supplementation.


La hemodiálisis, junto con la enfermedad renal crónica (ERC), intensifica el proceso inflamatorio y el estrés oxidativo en los pacientes en tratamiento. En este contexto, la suplementación con vitamina E puede mitigar los efectos nocivos resultantes de estos procesos. Esta es una revisión sistemática cuyo objetivo fue evaluar el efecto de la suplementación con vitamina E sobre biomarcadores inflamatorios y estrés oxidativo en pacientes con ERC en hemodiálisis. La revisión se elaboró según la metodología para revisiones sistemáticas y metanálisis (PRISMA) y la búsqueda se realizó en las bases de datos Pubmed, Cochrane Library, Scopus y Web of Science. El riesgo de sesgo de los estudios incluidos se evaluó con la Herramienta Cochrane de Riesgo de Sesgo (Cochrane Risk of Bias Tool). En esta revisión se incluyeron doce estudios desarrollados entre 2006 y 2020. La mayoría (n= 11) utilizó dosis de vitamina E que oscilaban entre 400 UI y 888 UI y el tiempo de suplementación osciló entre 2 semanas y 12 meses. De los 12 artículos incluidos, 25% (n= 3) analizaba la suplementación en biomarcadores inflamatorios, 50% (n= 6) en biomarcadores de estrés oxidativo y 25% (n= 3) en estos parámetros simultáneamente. Se observó un efecto positivo en 58,4% de los estudios (n= 7). Por lo tanto, la suplementación con vitamina E puede ser efectiva para mitigar el proceso inflamatorio y el estrés oxidativo, sin embargo, vale la pena señalar que el efecto depende de la dosis y el tiempo de suplementación.

ABSTRACT BACKGROUND: Despite the several options available for supplements containing vitamins C and E, evidence regarding the prevalence of deficiency or insufficiency of these vitamins is weak. OBJECTIVES: To estimate the prevalence of deficiency or insufficiency of vitamins C and E and associated factors among women of childbearing age, in Brazil. DESIGN AND SETTING: Systematic review and meta-analysis conducted at a Brazilian public university. METHODS: A search from index inception until May 2020 was conducted. Meta-analyses were performed using inverse variance for fixed models, with summary proportions calculation using Freeman-Tukey double arcsine (base case). Reporting and methodological quality were assessed using the Joanna Briggs Institute tool for prevalence studies. RESULTS: Our review identified 12 studies, comprising 1,316 participants, especially breastfeeding women. There was at least one quality weakness in all studies, mainly regarding sampling method (i.e. convenience sampling) and small sample size. The prevalence of vitamin C deficiency ranged from 0% to 40%. Only vitamin E deficiency was synthetized in meta-analyses, with mean prevalences of 6% regardless of the alpha-tocopherol cutoff in plasma, and 5% and 16% for cutoffs of < 1.6-12.0 mmol/l and < 16.2 mmol/l, respectively. The cumulative meta-analysis suggested that a trend to lower prevalence of vitamin E deficiency occurred in recent studies. CONCLUSIONS: Although the studies identified in this systematic review had poor methodological and reporting quality, mild-moderate vitamin C and E deficiencies were identified, especially in breastfeeding women. Thus, designing and implementing policies does not seem to be a priority, because the need has not been properly dimensioned among women of childbearing age in Brazil. ABSTRACT: The objective of this in vitro study was to assess the effects of two antioxidants (sodium ascorbate [SA] and alpha-tocopherol [AT]) on the adhesive interface and dentin bond strength immediately after bleaching with 38 % hydrogen peroxide (38HP) in endodontically-treated teeth. Two stages of experimentation were carried out. Bovine incisors were allocated into four groups (n = 10/group for each experiment): NB, non-bleached restored crowns; 38HP, bleached and immediately restored crowns; 38HP-SA, bleached crowns and SA use; and 38HP-AT, bleached crowns and AT use. Hybrid layer length in dentin (μm) and bond strength (MPa) were assessed with confocal microscopy laser and micro-shear bond strength (μSBS) test, respectively. Failure mode was determined by stereomicroscope. Data analysis was performed with analysis of variance (ANOVA) and Kruskal-Wallis, Dunn, Tukey, and Fisher-exact tests (a = 0.05). Higher values of hybrid layer length were observed similarly in the NB and 38HP-SA groups. The highest μSBS mean values were observed in the NB group (18 .51 ? 1.33), whereas the SBS values for 38HP-AT (1.68 ? 0.32) were similar to the 38HP group (1.61 ? 0.51) (p > 0.05) and significantly lower than the 38HP-SA group (5.78 ? 0.71). Adhesive failures were predominant in the 38HP and 38HP-AT groups. Cohesive and mixed failures were mostly observed in the NB and 38HP-SA groups, respectively. In conclusion, AT has no immediate effect on the hybrid layer formation and μSBS of dentin. Although SA promotes an increase in hybrid layer formation, it was not reflected in the μSBS values.


RESUMEN: El objetivo de este estudio in vitro fue evaluar los efectos de dos antioxidantes (ascorbato de sodio [AS] y alfa-tocoferol [AT]) sobre la interfaz adhesiva y la fuerza de unión de la dentina inmediatamente después del blanqueamiento con peróxido de hidrógeno al 38 % (38HP) en endodoncia. -Dientes tratados. Se llevaron a cabo dos etapas de experimentación. Los incisivos bovinos se dividieron en cuatro grupos (n = 10 / grupo para cada experimento): NB, coronas restauradas no blanqueadas; 38HP, coronas blanqueadas y restauradas inmediatamente; 38HP- AS, coronas blanqueadas y uso SA; y 38HP-AT, coronas blanqueadas y uso de AT. La longitud de la capa híbrida en dentina (μm) y la fuerza de unión (MPa) se evaluaron con láser de microscopía confocal y la prueba de fuerza de unión por micro-cizallamiento (μSBS), respectivamente. El modo de falla se determinó mediante estereomicroscopio. El análisis de los datos se realizó con análisis de varianza (ANOVA) y pruebas de Kruskal-Wallis, Dunn, Tukey y Fisher (α = 0,05). De manera similar, se observaron valores más altos de longitud de capa híbrida en los grupos NB y 38HP-AS. Los valores medios más altos de μSBS se observaron en el grupo NB (18,51 ? 1,33), mientras que los valores de SBS para 38HP-AT (1,68 ? 0,32) fueron similares a los del grupo 38HP (1,61 ? 0,51) (p> 0,05) y significativamente más bajos que el grupo 38HP-AS (5,78 ? 0,71). Las fallas adhesivas fueron predominantes en los grupos de 38HP y 38HP-AT. Las fallas cohesivas y mixtas se observaron principalmente en los grupos NB y 38HP-AS, respectivamente. En conclusión, la AT no tiene un efecto inmediato sobre la formación de la capa híbrida y el μSBS de dentina. Aunque AS promueve un aumento en la formación de capas híbridas, no se refleja en los valores de μSBS.

ABSTRACT

In the present study, the effect of vitamin E supplementation 450 mg/kg diet was appraised in the process of induced wound healing in Nile tilapias Oreochromis niloticus. Fish were distributed into 18 tanks (10 fish each). Nine tanks were fed the non-supplemented diet and the other 9 tanks were fed 450 mg vitamin E for 60 days. Subsequently, the fish were anesthetized and the epidermis and dermis were surgically removed. The rate of cicatricial retraction and appearance of the wounds, and the histomorphometry of mucous cells, chromatophores, revascularization, inflammatory cells, presence of fibroblasts, collagen fibers, and scales were checked after 3-, 7-, 14-, 21-, and 28-days post-wounding. The retraction rate of the wound was significantly higher in the supplemented fish. The higher concentrations of inflammatory cells, mucous cells, and chromatophores, as well as the production and organization of collagen fibers, resulted in a higher retraction rate. We concluded that a dietary supplementation diet improves specific aspects of the cutaneous healing process in Nile tilapia fish.(AU)


No presente estudo, o efeito da suplementação com vitamina E de 450 mg / kg de dieta foi avaliado no processo de cicatrização induzida de feridas em tilápias do Nilo, Oreochromis niloticus. Os peixes foram distribuídos em 18 tanques (N=10), sendo 9 tanques com dieta não suplementada e os outros 9 tanques suplementados com 450 mg de vitamina E por 60 dias. Posteriormente, os peixes foram anestesiados e a epiderme e derme foram removidas cirurgicamente. Nos tempos pré-determinado de 3, 7, 14, 21 e 28 dias após a ferida foi analisado a taxa de retração cicatricial, a aparência das feridas e a histomorfometria das células mucosas, dos cromatóforos, das células inflamatórias, a revascularização, a presença de fibroblastos, de fibras de colágeno e escamas. A taxa de retração da ferida foi significativamente maior nos peixes suplementados. As maiores concentrações de células inflamatórias, mucosas e cromatóforos, bem como a produção e organização das fibras de colágeno, resultaram em uma maior taxa de retração. Concluímos que a dieta de suplementação melhora aspectos específicos do processo de cicatrização cutânea em peixes de tilápia do Nilo.(AU)

Resumo Os triglicerídeos são um dos componentes lipídicos do sangue e sua elevação é fator de risco maior para doenças cardiovasculares. A prática de exercícios é responsável pela diminuição da gordura presente no organismo, além de equilibrar a quantidade de energia ingerida e utilizada. A vitamina "E" é um antioxidante lipossolúvel tecidual que sustenta a cadeia de peroxidação lipídica, prevenindo sua oxidação, podendo atuar na redução da trigliceridemia. Este estudo teve como objetivo avaliar as concentrações plasmáticas de triglicerídeos de equinos após serem submetidos ao esforço físico e suplementados com vitamina "E". Para tanto, foram utilizados dez cavalos que realizaram exercício de longa duração em esteira em duas etapas, a primeira sem suplementação e a segunda com suplementação de vitamina "E". Coletas de sangue foram realizadas nas duas fases, antes do teste (M0), imediatamente após o exercício (PE) e nos tempos 6, 12, 24, 48, 72 e 96 horas subsequentes. Observou-se aumento na concentração sérica de triglicerídeos em ambas as fases, sendo os valores da segunda superiores aos da primeira. Não se evidenciou neste estudo, portanto, redução na trigliceridemia em equinos suplementados com vitamina E.


Resumen Los triglicéridos son uno de los componentes lipídicos de la sangre y su elevación es un factor de riesgo mayor para las enfermedades cardiovasculares. La práctica de ejercicios es responsable de la disminución de la grasa presente en el organismo, además de equilibrar la cantidad de energía ingerida y utilizada. La vitamina E es un antioxidante liposoluble tecidual que sostiene la cadena de peroxidación lipídica, lo que previene su oxidación, y de este modo puede actuar en la reducción de la trigliceridemia. Este estudio tuvo como objetivo evaluar las concentraciones plasmáticas de triglicéridos de equinos después de ser sometidos al esfuerzo físico y suplementados con vitamina E. Para ello se utilizaron diez caballos que realizaron ejercicio de larga duración en cinta en dos etapas: la primera, sin suplementación; y la segunda, con suplementación de vitamina E. Las colectas de sangre se realizaron en las dos fases, antes de la prueba (M0), inmediatamente después del ejercicio (PE) y en los tiempos 6, 12, 24, 48, 72, 96 y 96 horas. Se observó un aumento en la concentración sérica de triglicéridos en ambas fases, siendo los valores de la segunda superiores a los de la primera. No se evidenció en este estudio, reducción en la trigliceridemia en equinos suplementados con vitamina E.


Abstract Triglycerides are one of the blood lipid components and their increasing values are a risk factor for cardiovascular diseases. Working out helps to reduce fat in the body as well as to balance the amount of energy uptake vs. energy use. Vitamin E is a liposoluble antioxidant tissue that supports the lipid peroxidation chain, thus preventing the oxidation and, in turn, acting as a triglyceridemia reducing agent. This study aims to evaluate the serum concentrations of triglycerides in Equi after doing a physical effort and receiving vitamin E. The study included ten horses that did a long-run workout session in two rounds: the first one without receiving any supplement; the second one, receiving vitamin E supplements. Blood samples were collected in the rounds: one before the test (M0), immediately after the workout (PE) and at the 6, 12, 24, 48, 72, 96 and 96 hours. An increase was observed in the serum concentration of triglycerides in both rounds, with higher values in the second round. The study did not find any reduction in the triglyceridemia among Equi receiving vitamin E supplements.

Objective: Vital bleaching is a popular treatment option for discolored teeth; but at post-treatment stage, loss of adhesion is highly reported. Literature focused on antioxidant application for the answer of this issue. The aim of this study was to compare the effects of six different antioxidants on color stability of bleached teeth. Material and Methods: This study included total of 84 extracted intact non-carious lower incisors. 35% hydrogen peroxide was applied on the labial surfaces of specimens in accordance with manufacturer's instructions. The bleached teeth were divided into 7 groups. No antioxidants were applied to the control group. For the experimental groups, the following antioxidants were applied for 10 minutes each: 5% proanthocyanidin, 5% sodium ascorbate, 5% lycopene, %5 green tea, %5 white tea and %5 α-tocopherol. CIE L*, a* and b* values of the teeth were measured by a spectrophotometer. One-way ANOVA was used to determine the differences among the groups. Multiple comparisons were examined with Tukey HSD. Results: The one-way ANOVA test revealed a statistically significant difference between the groups (p < 0.005). Highest color change was observed in lycopene group and the lowest in green tea group. Conclusion: Proanthocyanidin, white tea and green tea could be considered as post-bleaching antioxidant alternatives based on their herbal nature. (AU)


Objetivo: O clareamento vital é uma opção popular de tratamento para dentes descoloridos, mas na fase pós-tratamento, a perda de adesão é altamente relatada. A literatura enfocou a aplicação de antioxidantes para a resposta desta questão. O objetivo deste estudo foi comparar os efeitos de seis diferentes antioxidantes na estabilidade da cor de dentes clareados. Material e Métodos: Este estudo incluiu um total de 84 incisivos inferiores extraídos, intactos e não cariados. Peróxido de hidrogênio a 35% foi aplicado nas superfícies labiais dos espécimes de acordo com as instruções do fabricante. Os dentes clareados foram divididos em 7 grupos. Nenhum antioxidante foi aplicado ao grupo controle. Para os grupos experimentais, os seguintes antioxidantes foram aplicados por 10 minutos cada: proantocianidina a 5%, ascorbato de sódio a 5%, licopeno a 5%, chá verde a 5%, chá branco a 5% e α-tocoferol a 5%. Os valores CIE L *, a * e b * dos dentes foram medidos por um espectrofotômetro. ANOVA um fator foi usada para determinar as diferenças entre os grupos. As comparações múltiplas foram examinadas com Tukey HSD. Resultados: O teste ANOVA revelou uma diferença estatisticamente significativa entre os grupos (p <0,005). A maior mudança de cor foi observada no grupo do licopeno e a menor no grupo do chá verde. Conclusão: Proantocianidina, chá branco e chá verde podem ser considerados como alternativas antioxidantes pós-clareamento com base em sua natureza fitoterápica. (AU)

ABSTRACT

The present study was planned to determine the effect of Epigallocatechin Gallate (EGCG), Alpha tocopherol and their combination as an antioxidant in TCM-199 media for in vitro maturation (IVM) and in vitro fertilization (IVF) in bovine oocytes. Cumulus-oocyte complexes (COCs) were aspirated from the ovaries derived from slaughter house and in vitro cultured in three groups using TCM-199 supplemented with EGCG @10 μM, Alpha tocopherol @100 μM, and Combination (EGCG @10 μM plus Alpha tocopherol @100 μM). Oocytes of a control group were matured in TCM-199 medium without any treatment. After IVM, cumulus-free oocytes were co-incubated with frozen-thawed spermatozoa for 15–18 h. Compared to no addition, the presence of EGCG @10 μM in medium during IVM significantly (p<0.05) increased the proportion of maturation and fertilization rate. Combination produced significantly higher percentage of in vitro matured bovine oocytes compared to the alpha tocopherol @100 μM alone. The results suggest that EGCG @10 μM in IVM medium had better effect than Alpha tocopherol alone and Combination on in vitro maturation and subsequent fertilization of bovine oocytes.

ABSTRACT Objective: To determine the concentration of alpha-tocopherol in umbilical cord serum of full-term and preterm newborns, in order to assess the nutritional status of both groups in relation to the vitamin and its possible correlation with intrauterine growth. Methods: A cross-sectional observational study conducted with 140 newborns, of which 64 were preterm and 76 were full-term. They did not have any malformations, they came from healthy mothers, who were nonsmokers, and delivered a single baby. Intrauterine growth was evaluated by weight-to-gestational age at birth, using Intergrowth-21st. Thealpha-tocopherol levels of umbilical cord serum were analyzed by High Performance Liquid Chromatography. Results: The mean concentration of alpha-tocopherol in umbilical cord serum for preterm and full-term infants was 263.3±129.5 and 247.0±147.6 µg/dL (p=0.494). In the preterm group, 23% were small for gestational age, whereas in the full-term group, this percentage was only 7% (p=0.017). Low levels of vitamin E were found in 95.3% of preterm infants and 92.1% of full-term infants. There was no correlation between alpha-tocopherol levels and weight to gestational age Z score (p=0.951). Conclusions: No association was found between alpha-tocopherol levels and weight to gestational age at birth. Intrauterine growth restriction was more frequent in preterm infants and most infants had low levels of vitamin E at the time of delivery.


RESUMO Objetivo: Determinar a concentração de alfatocoferol em soro de cordão umbilical de recém-nascidos a termo e pré-termo, a fim de avaliar o estado nutricional de ambos os grupos com relação a essa vitamina e sua possível correlação sobre o crescimento intrauterino. Métodos: Estudo observacional de caráter transversal realizado com 140 recém-nascidos, 64 pré-termo e 76 a termo, sem malformações, oriundos de mães saudáveis, não fumantes e com parto de concepto único. O crescimento intrauterino foi avaliado pelo índice peso por idade gestacional ao nascer, utilizando a Intergrowth-21st. Os níveis de alfatocoferol do soro do cordão umbilical foram analisados por cromatografia líquida de alta eficiência. Resultados: A concentração média de alfatocoferol no soro do cordão umbilical para recém-nascidos pré-termo e a termo foi de, respectivamente, 263,3±129,5 e 247,0±147,6 µg/dL (p=0,494). Baixos níveis de vitamina E foram encontrados em 95,3% dos prematuros e em 92,1% dos neonatos a termo. Nogrupo pré-termo, 23% eram pequenos para a idade gestacional, enquanto no grupo a termo esse percentual foi de apenas 7% (p=0,017). Não houve correlação entre os níveis de alfatocoferol e o escore Z de peso para idade gestacional (p=0,951). Conclusões: Não foi encontrada associação entre os níveis de alfatocoferol e a adequação do peso à idade gestacional ao nascer. A restrição do crescimento intrauterino foi mais frequente nos nascidos pré-termo, e a maioria dos recém-nascidos apresentou níveis baixos de vitamina E no momento do parto.

Oxidative stress and inflammation are the key players in the development of motor dysfunction post-spinal cord ischemic reperfusion injury (SC-IRI). This study investigated the protective effect of concomitant pre-administration of melatonin and alpha-tocopherol on the early complications (after 48 hours) of spinal cord IRI injury in rats. Melatonin or α-tocopherol were preadministered either individually or in combination for 2 weeks, then rats were exposed SC-IRI. Neurological examinations of the hind limbs and various biochemical markers of oxidative stress and inflammation in the SC tissue were assessed. Solely pre-administration of either melanin or α-tocopherol significantly but partially improved motor and sensory function of the hind limbs mediated by partial decreases in SC levels of MDA, AOPP and PGE2 levels and activities of SOD, partial significant decreases in plasma levels of total nitrate/nitrite and significant increases in AC activity of GSH-Px. However, combination therapy of both drugs resulted in the maximum improvements in all neurological assessments tested and biochemical endpoints. In conclusion, by their synergistic antioxidant and antiinflammatory actions, the combination therapy of melatonin and α-tocopherol alleviates SC-IRI induced paraplegia.


El estrés oxidativo y la inflamación son claves en el desarrollo de la disfunción motora posterior a lesión isquémica de la médula espinal (SC-IRI). Este estudio investigó acerca del efecto protector de la administración previa concomitante de la melatonina y alfa-tocoferol en las complicaciones tempranas (después de 48 horas) de la lesión de IRI de la médula espinal en ratas. La melatonina o el α-tocoferol se administraron individualmente o en combinación durante 2 semanas, luego las ratas fueron expuestas a SC-IRI. Se evaluaron los exámenes neurológicos de las miembros pélvicos y diversos marcadores bioquímicos de estrés oxidativo e inflamación en el tejido subcutáneo. Solo la administración previa de melatonina o α-tocoferol mejoró parcial y significativamente la función motora y sensorial de los miembros pélvicos mediadas por disminuciones parciales en los niveles de SC de los niveles de MDA, AOPP y PGE2 y las actividades de la SOD, disminuciones significativas parciales en los niveles plasmáticos del total nitrato / nitrito y aumentos significativos en la actividad de AC de GSH-Px. Sin embargo, se observaron los mejores resultados durante la combinación de ambos fármacos en todas las evaluaciones neurológicas y en los puntos finales bioquímicos. En conclusión, debido a sus acciones antioxidantes y antiinflamatorias sinérgicas, la terapia de melatonina y α-tocoferol alivia la paraplejía inducida por SC-IRI.

RESUMO Objetivos: Determinar a concentração de alfa-tocoferol no leite materno em diferentes períodos de lactação e estimar o provável fornecimento de vitamina E ao lactente. Métodos: Estudo longitudinal observacional realizado com 100 puérperas atendidas para o parto no Hospital Universitário Ana Bezerra (HUAB) da Universidade Federal do Rio Grande do Norte (UFRN), em Santa Cruz (RN). Foram coletados leite colostro (n=100), leite de transição (n=77) e leite maduro (n=63) no seguimento da lactação. O alfa-tocoferol foi analisado por cromatografia líquida de alta eficiência. O fornecimento de vitamina E para o neonato foi estimado comparando-se o requerimento nutricional de vitamina E (4 mg/dia) com a ingestão diária de leite. Resultados: A concentração média de alfa-tocoferol encontrada nos leites colostro, de transição e maduro foi 40,5±15,0 µmol/L, 13,9±5,2 µmol/L e 8,0±3,8 µmol/L, respectivamente (p<0,001). A possível ingestão desses leites pelo lactente forneceu 6,2 mg/dia de vitamina E no colostro, 4,7 mg/dia no de transição e 2,7 mg/dia no maduro (p<0,0001), evidenciando que apenas o último não garantiu a quantidade recomendada dessa vitamina. Conclusões: Os níveis de alfa-tocoferol no leite diminuíram com a progressão da lactação, e a provável ingestão dos leites colostro e de transição conseguiu atender ao requerimento nutricional do lactente. O leite maduro pode fornecer menores quantidades da vitamina E, o que torna importante o estudo dos fatores que se associam a esses baixos níveis.


ABSTRACT Objectives: To determine the alpha-tocopherol concentration in breast milk at different periods of lactation and to estimate the possible supply of vitamin E to the infant. Methods: A longitudinal observational study was carried out with 100 mothers at University Hospital Ana Bezerra (HUAB), at Universidade Federal do Rio Grande do Norte, in Santa Cruz (RN), Northeast Brazil. Samples of colostrum (n=100), transitional milk (n=77), and mature milk (n=63) were collected. Alpha-tocopherol was analyzed by high-performance liquid chromatography. Vitamin supply to the newborn was estimated by comparing the nutritional requirement of vitamin E (4 mg/day) with the potential daily intake of milk. Results: The mean alpha-tocopherol concentration found in colostrum, transitional, and mature milk was 40.5±15.0 µmol/L, 13.9±5.2 µmol/L, and 8.0±3.8 µmol/L, respectively (p<0.001). The possible effect of these milks offered to the infant 6.2 mg/day of vitamin E in colostrum, 4.7 mg/day in transitional milk, and 2.7 mg/day in mature milk (p<0.0001), shows that only the mature milk did not guarantee the recommended quantity of this vitamin. Conclusions: Alpha-tocopherol levels in human milk decrease through the progression of lactation, and the possible intake of colostrum and transitional milk met the nutritional requirement of the infant. Mature milk may provide smaller amounts of vitamin E. Thus, it is important to study the factors that are associated with such low levels.

Abstract: Objective: To evaluate the effect of maternal supplementation with vitamin E on the concentration of α-tocopherol in colostrum and its supply to the newborn. Method: This randomized clinical trial enrolled 99 healthy adult pregnant women; of these, 39 were assigned to the control group and 60 to the supplemented group. After an overnight fast, 5 mL of blood and 2 mL of colostrum were collected. After the first sampling (0 h milk), the supplemented group received 400 IU of supplementary vitamin E. Another 2 mL milk aliquot was collected in both groups 24 h after supplementation (24 h milk). The samples were analyzed by high-performance liquid chromatography. The α-tocopherol content provided by colostrum was calculated by considering a daily intake of 396 mL of milk and comparing the resulting value to the recommended daily intake for infants aged 0-6 months (4 mg/day). Results: The initial mean concentration of α-tocopherol in colostrum was 1509.3 ± 793.7 µg/dL in the control group and 1452.9 ± 808.6 µg/dL in the supplemented group. After 24 h, the mean α-tocopherol concentration was 1650.6 ± 968.7 µg/dL in the control group (p > 0.05) and 2346.9 ± 1203.2 µg/dL in the supplemented group (p < 0.001), increasing the vitamin E supply to the newborn to 9.3 mg/day. Initially, 18 women in the supplemented group provided colostrum α-tocopherol contents below 4 mg/day; after supplementation only six continued to provide less than the recommended amount. Conclusion: Maternal vitamin E supplementation increases the supply of the vitamin to the infant by providing more than twice the Recommended Daily Intake.


Resumo: Objetivo: Avaliar o efeito da suplementação materna com vitamina E sobre a concentração de α-tocoferol no colostro e o fornecimento dessa para o recém-nascido. Método: O estudo clínico randomizado foi feito com 99 parturientes adultas e saudáveis, 39 alocadas no grupo controle e 60 no grupo suplementado. Após jejum noturno, foram coletados 5 mL de sangue e 2 mL de colostro das parturientes. Após a primeira coleta (leite 0 h), o grupo suplementado recebeu suplementação com 400 UI de vitamina E. Foi feita nova coleta de 2 mL de colostro, em ambos os grupos, 24 h após a suplementação (leite 24 h). As amostras foram analisadas por cromatografia líquida de alta eficiência. A quantidade de α-tocoferol fornecida pelo colostro foi considerada para uma ingestão diária de 396 mL de leite e comparada com a ingestão diária recomendada para crianças de 0 a 6 meses (4 mg/dia). Resultados: A concentração média inicial de α-tocoferol no colostro foi de 1.509,3 ± 793,7 µg/dL no grupo controle e 1.452,9 ± 808,6 µg/dL no grupo suplementado. Após 24 horas a concentração média de α-tocoferol no grupo controle foi de 1.650,6 ± 968,7 µg/dL (p > 0,05) e de 2.346,9 ± 1203,2 µg/dL (p < 0,001) no grupo suplementado. Aumentou-se assim a oferta de vitamina E para o recém-nascido para 9,3 mg/dia. Inicialmente 18 mulheres do grupo suplementado forneciam valores inferiores a 4 mg/dia de α-tocoferol em seu colostro, após suplementação apenas seis continuaram a fornecer quantidade inferior ao recomendado. Conclusão: A suplementação materna com vitamina E promove o aumento do fornecimento da vitamina para o recém-nascido e fornece mais do que o dobro da ingestão diária recomendada.

Aim: To examine the effects of ascorbic acid and alpha-tocopherol on the locomotor functional recovery of rats with incomplete spinal cord injury. Methods: A total of 80 male Sprague-Dawley rats (180–200 g) were used. Seventy of them were subjected to spinal cord injury (SCI) and given various doses of ascorbic acid and alpha-tocopherol as treatment for 84 days (12 weeks). The remaining 10 rats were used as a control (without SCI, untreated). Basso, Beattie, and Bresnahan (BBB) and Tarlov scores were used to assess the locomotor recovery. Results: Compared with the baseline value, all the rat groups tested (i.e. 3, 4, 5, 6, 7 and 8) showed gradual improvements in the BBB and Tarlov scores at the end of the first week. These improvements were sustained at all-time points until the completion of the trial period. Besides, when compared with disease control, positive improvements were observed in all the groups tested with respect to their BBB and Tarlov scores throughout the trial period. When the low-dose ascorbic acid and alpha-tocopherol groups were compared with the high-dose ascorbic acid and alpha-tocopherol groups, positive differences were observed in the BBB and Tarlov scores. When compared with the ascorbic acid treatment group significant additional improvements were observed in the alpha-tocopherol groups, especially in the high-dose alpha-tocopherol group. Conclusion: Administration of high-dose alpha-tocopherol enhances the SCI-induced locomotor functional recovery as it is more effective than the ascorbic acid.

O alfa- e o gama-tocoferol estão entre os homólogos da vitamina E, que possui importante papel como antioxidante. As fontes dietéticas mais ricas em vitamina E são os óleos vegetais. Este trabalho avaliou os níveis de alfa- e gama-tocoferol nos óleos de canola, girassol, milho e soja, e averiguou sua variação com o tempo de armazenamento. Os óleos vegetais foram adquiridos nos supermercados da cidade de Natal/RN, e mantidos sob temperatura ambiente e ao abrigo da luz. As análises foram realizadas em diferentes momentos durante o armazenamento (tempo 0 e após 30, 60 e 90 dias). Cromatografia líquida de alta eficiência (CLAE) foi usada para determinar as concentrações dos analitos. A análise estatística foi realizada utilizando-se o teste de variância (ANOVA) e as diferenças foram consideradas estatisticamente significativas quando p < 0,05. Ao final do tempo de armazenamento (90 dias) foi verificada diminuição significativa nos níveis de alfa- e gama-tocoferolde 38,7 % e 36,0 %, no óleo de canola; 42,2 % e 22,2 %, no óleo de soja; 28,3 % e 29,2 %, no óleo de girassol;39,0 % e 17,9 %, no óleo de milho, respectivamente. As concentrações de alfa- e gama-tocoferol nos óleos vegetais reduziram significativamente após armazenamento, sob as condições empregadas no estudo.


The alpha- and gamma-tocopherol are among the homologues of vitamin E which plays a key role as anantioxidant. The richest dietary sources of vitamin E are vegetable oils. This study aimed at evaluating thealpha- and gamma-tocopherol contents in oils from canola, sunflower, corn and soybeans, and to verify their variations after storage time. The vegetable oils were purchased at supermarkets in Natal / RN, and they were keptat room temperature and protected from light. The tests was made at different times during storage (time 0 and after 30, 60 and 90 days). The high performance liquid chromatography (HPLC) was used to determinetheir concentrations. The statistical analysis was performed using analysis of variance (ANOVA) test and thedifferences were considered statistically significant when p < 0.05. By the end of the storage time (90 days), thealpha- and gamma-tocopherol contents showed a significant decrease of 38.7 % and 36.0 % for canola oil; 42.2 %and 22.2 % in soybean oil; 28.3 % and 29.2 % in sunflower oil; 39.0 % and 17.9 % in corn oil, respectively.The concentrations of alpha- and gamma-tocopherol in vegetable oils significantly decreased after the storageperiod under the conditions used in this study.

BACKGROUND/OBJECTIVES: Vitamin E is a fat-soluble vitamin and functions primarily as a lipid antioxidant. Inadequate vitamin E status may increase risk of several chronic diseases. Thus, the objectives of this study were to estimate intake and plasma concentration of each tocopherol and to evaluate vitamin E status of Korean adults. SUBJECTS/METHODS: Three consecutive 24-h food recalls and fasting blood samples were collected from healthy 20- to 59-y-old adults (33 males and 73 females) living in the Seoul metropolitan area, South Korea. alpha-, beta-, delta-, and gamma-tocopherol intakes and plasma concentrations of tocopherols (alpha-, delta-, and gamma-tocopherol) were analyzed by gender. RESULTS: Dietary vitamin E and total vitamin E intake (dietary plus supplemental vitamin E) was 17.68 +/- 14.34 and 19.55 +/- 15.78 mg alpha-tocopherol equivalents, respectively. The mean daily alpha-tocopherol, and gamma-tocopherol intakes were 3.07 +/- 2.27 mg and 5.98 +/- 3.74 mg, respectively. Intakes of total vitamin E and each tocopherol of males were significantly higher than those of females (P or = 0.05). Plasma alpha-tocopherol was negatively correlated with gamma-tocopherol intake (P < 0.05). Twenty-three percent of the subjects had plasma alpha-tocopherol concentrations < 12 micromol/L indicating a biochemical deficiency of vitamin E. Approximately 8% and 9% of these participants had plasma alpha-tocopherol:total lipid ratio less than 1.59 micromol/mmol and plasma alpha-tocopherol:total cholesterol ratio less than 2.22 micromol/mmol, respectively, which are also indicative of vitamin E deficiency. CONCLUSIONS: Vitamin E intakes of Korean adults were generally adequate with the Korean Dietary Reference Intakes for vitamin E. However, alpha-tocopherol intake was lower than that reported in other countries, and 23% of the subjects in the current study were vitamin E deficient based on plasma alpha-tocopherol concentrations.

ABSTRACT

OBJECTIVES: Some antioxidants are believed to restore dentin bond strength after dental bleaching. This study was done to evaluate the influence of antioxidants on the bond strength of bleached bovine dentin. MATERIALS AND METHODS: Thirty incisors were randomly assigned to 10 groups (two unbleached control and eight bleached groups: immediate bonding IB, 4 wk delayed bonding DB, 10% sodium ascorbate treated SA, 10% alpha-tocopherol treated TP groups). Teeth in half of groups were subjected to thermal stress, whereas the remaining groups were not. Resin-dentin rods with a cross-sectional area of 2.25 mm2 were obtained and microtensile bond strength was determined at a crosshead speed of 1 mm/min. Fifteen specimens were prepared for SEM to compare the surface characteristics of each group. The change in dentin bond strength from thermal stress and antioxidant treatment was evaluated using two-way analysis of variance (ANOVA) and Sheffe's post hoc test at a significance level of 95%. RESULTS: The control group exhibited the highest bond strength values, whereas IB group showed the lowest value before and after thermocycling. The DB group recovered its bond strength similar to that of the control group. The SA and TP groups exhibited similar bond strength values with those of the control and DB groups before thermocycling. However, The TP group did not maintain bond strength with thermal stress, whereas the SA group did. CONCLUSIONS: Applying a 10% sodium ascorbate solution rather than 10% alpha-tocopherol solution for 60 sec is recommended to maintain dentin bond strength when restoring non-vitally bleached teeth.

Objective: To systematize information about vitamin E concentration in human milk and the variables associated with this composition in order to find possible causes of deficiency, supporting strategies to prevent it in postpartum women and infants. Source: Studies published between 2004 and 2014 that assayed alpha-tocopherol in human milk of healthy women by high performance liquid chromatography were evaluated. The keywords used were "vitamin E", "alpha-tocopherol", "milk, human", "lactation", and equivalents in Portuguese, in the BIREME, CAPES, PubMed, SciELO, ISI Web of Knowledge, HighWire Press, Ingenta, and Brazilian Digital Library of Theses and Dissertations databases. Summary of the findings: Of the 41 publications found on the subject, 25 whose full text was available and met the inclusion criteria were selected. The alpha-tocopherol concentrations found in milk were similar in most populations studied. The variable phase of lactation was shown to influence vitamin E content in milk, which is reduced until the mature milk appears. Maternal variables parity, anthropometric nutritional status, socioeconomic status, and habitual dietary intake did not appear to affect the alpha-tocopherol levels in milk. However, the influence of the variables maternal age, gestational age, biochemical nutritional status in alpha-tocopherol, and maternal supplementation with vitamin E had conflicting results in the literature. Conclusion: Alpha-tocopherol concentration in milk decreases during lactation, until the mature milk appears. To confirm the influence of some maternal and child variables on milk vitamin E content, further studies with adequate design are needed. .


Objetivo: Sistematizar informações sobre a concentração de vitamina E no leite humano e variáveis associadas a essa composição, a fim de encontrar possíveis causas de deficiência, subsidiando estratégias para prevenção desta em puérperas e lactentes. Fonte dos dados: Foram avaliados estudos publicados entre 2004 e 2014 que determinaram o alfa-tocoferol no leite humano de mulheres saudáveis por Cromatografia Líquida de Alta Eficiência. Os descritores utilizados foram "leite humano", "alfa-tocoferol", "vitamina E", "lactação" e seus equivalentes em inglês, nas bases de dados BIREME, Periódicos CAPES, Pub-Med, SciELO, ISI Web of Knowledge, HighWire Press, Ingenta e Biblioteca Digital Brasileira de Teses e Dissertações. Síntese dos dados: Das 41 publicações encontradas sobre a temática, 25 foram selecionadas, por possuirem texto completo disponível e se encaixarem nos critérios de inclusão. As concentrações encontradas de alfa-tocoferol no leite foram semelhantes na maioria das populações estudadas. A variável fase de lactação mostrou influenciar o conteúdo dessa vitamina no leite, que vai sendo reduzido até o leite maduro. As variáveis maternas paridade, estado nutricional antropométrico, condição socioeconômica e ingestão dietética habitual parecem não afetar os níveis de alfa-tocoferol no leite. Entretanto, a influência das variáveis idade materna, idade gestacional, estado nutricional bioquímico em alfa-tocoferol e suplementação materna com vitamina E possui resultados conflitantes na literatura. OBJETIVO: Comparar a concentração de alfa-tocoferol no soro entre puérperas atendidas em maternidades das redes pública e privada da cidade de Natal (RN), Brasil. MÉTODOS: Participaram do estudo 209 puérperas, sendo 96 mulheres provenientes da rede privada e 113, da rede pública, entre 24 e 48 horas pós-parto. Foram incluídas no estudo parturientes com idade a partir de 12 anos, sem patologias associadas à gestação, que tiveram concepto único sem má-formação. Mulheres descompensadas clinicamente e com gestação múltipla foram excluídas. Amostras de 5 mL de sangue de cada participante foram coletadas no período de jejum, antes da primeira refeição do dia. A concentração de alfa-tocoferol no soro (µg/dL) foi determinada por cromatografia líquida de alta eficiência (CLAE). A diferença estatística entre as médias foi testada utilizando o teste t de Student. RESULTADOS: As médias da concentração de alfa-tocoferol nas puérperas da rede pública e da rede privada, respectivamente, foram 1.115,7±341,4 μg/dL e 1.355,7±397,6 μg/dL, tendo sido observada diferença estatisticamente significante (p=0,000687). Para avaliação individual da concentração de vitamina E, a deficiência foi identificada quando alfa-tocoferol <11,6 μmol/L ou <499,6 µg/dL. As puérperas da rede pública tiveram um percentual de 5,3% (n=6) de deficiência, enquanto nas participantes da rede privada tal carência não foi encontrada. Todavia, baixas concentrações (alfa-tocoferol entre 11,6-16,2 μmol/L ou 499,6-697,7 µg/dL) ocorreram tanto na rede pública como na privada: 9,7% (n=11) e 4,2% (n=4), respectivamente. PURPOSE: To compare the concentration of serum alpha-tocopherol during the postpartum period in women admitted to public and private hospitals in Natal (RN), Brazil. METHODS: The study included 209 women in the postpartum period, 96 of them from private hospitals and 113 from public hospitals, studied between 24 and 48 hours postpartum. RESULTS: The mean concentration of alpha-tocopherol was 1.115.7 µg/dL in puerperae from the public network and 1.355.7±397.6 µg/dL in puerperae from the privte network , with a significant difference between groups (p=0.000687). Vitamin E concentration was determined individually and an alpha-tocopherol level <11.6 µmol/L or <499.6 µg/dL was considered to indicate deficiency. Vitamin E deficiency was detected in 5.3% of puerperae from the public network (n=6), whereas no deficiency was detected among women from the private network. However, low concentrations of alpha-tocopherol (11.6 to 16.2 µmol/L or 499.6 to 697.7 µg/dL) was detected in both groups, i.e., in 9.7% of the women from the public network (n=11) and in 4.2% for the women from the private network (n=4). CONCLUSION: These results highlight that women assisted in the public sector were more vulnerable to developing low concentrations of alpha-tocopherol than women assisted in the private sector. .

ABSTRACT

Poco se sabe sobre los cambios en la actividad de las enzimas séricas relacionadas con la función hepática durante la hipervitaminosis E. En el presente trabajo se estudió el efecto de la administración intraperitoneal de 50, 100, 200 y 400 mg de vitamina E/día, durante 20 días sobre la actividad enzimática sérica en 60 ratas Wistar machos, de 12 semanas de edad, con pesos entre 180 y 200 gramos. El grupo control estuvo integrado por 15 ratas Wistar sanas, con edad y peso similares a los animales tratados. Al final del estudio, se tomaron muestras de sangre para la determinación de la vitamina E y la actividad de las enzimas: alanina aminotransferasa (ALT), aspartato aminotransferasa (AST), α-amilasa (AMS), arginasa (ARG), fosfohexosaisomerasa (PHI), fosfatasa alcalina (ALP), γ-glutamiltransferasa (γ-GT) y 5´-nucleotidasa (5´-N). La administración de vitamina E en exceso incrementó de manera significativa (p<0,05) el contenido sérico de la vitamina E y la actividad de todas las enzimas valoradas (p< 0,05); mientras que la α-amilasa disminuyó (p< 0,05) al ser comparada con los controles no tratados. Nuestros resultados proporcionan evidencia que la administración a corto plazo de dosis altas de vitamina E, produce un incremento en la actividad de las enzimas marcadoras de daño hepático (como aminotransferasas, ARG y PHI) y de colestasis (como ALP, 5´-N y γ-GT), que se corresponde con la forma mixta de enfermedad hepática (daño+colestasis).


Little is known about the possible changes in blood enzyme activity related to liver function during hypervitaminosis E. In the present work the effects of intraperitoneal administration of 50, 100, 200 and 400 mg of vitamin E (α-tocopherol) daily for 20 days, respectively, on the serum enzyme activity in 60 white male Wistar rats, aged 12 weeks and weighing 180-200 g, were studied. The group control was integrated by 15 healthy rats with similar characteristics (age and weight) to treated animals. Excess of vitamin E produced a significant (p<0.05) increase in the serum content of vitamin E and in the activity (p<0.05) of the following enzymes: alanine aminotransferase (ALT), aspartate aminotransferase (AST), arginase (ARG), phosphohexosaisomerase (PHI), alkaline phosphatase (ALP), γ-glutamyltransferase (γ-GT) and 5´-nucleotidase (5´-N) while α-amylase (AMS) decreased (p<0.05) on comparing with the control group. These changes depend on the doses given of vitamin E. In conclusion, our results provide evidence that short-term administration of high doses of vitamin E produces an increase in the activity of the enzymes marker of liver damage (as aminotransferases, ARG and PHI) and of cholestasis (as ALP, γ-GT and 5´-N) that correspond to the mixed form of liver disease (injury+cholestasis).

To evaluate and compare the levels of α-tocopherol in colostrum and in the serum of healthy and diabetic mothers. METHODS: This cross-sectional study enrolled 51 volunteer mothers, 20 with the diagnosis of gestational diabetes mellitus and 31 without associated diseases. Serum and colostrum samples were collected in fasting in the immediate postpartum period and α-tocopherol was analyzed by high performance liquid chromatography (HPLC). In order to define the nutritional status of vitamin E, the cutoff point for the serum (697.7µg/dL) was adopted. Student's t-test for independent variables compared the average concentrations of α-tocopherol in the serum and in the colostrum between control and gestational diabetes mellitus groups. Pearson's correlation was used to assess the relationship between the concentration of α-tocopherol in serum and colostrum for both groups. Differences were considered significant when p<0.05. RESULTS: The α-tocopherol concentration in colostrum was 1,483.1±533.8µg/dL for Control Group and 1,368.8±681.8µg/dL for diabetic women, without differences between groups (p=0.50). However, α-tocopherol concentration in the serum was 1,059.5±372.7µg/dL in the Control Group and 1,391.4±531.5µg/dL in the diabetic one (p<0.01). No correlation was found between the concentration of α-tocopherol in the serum and in the colostrum for control and diabetic groups. Evaluar y comparar la concentración de α-tocoferol en la leche calostro y en el suero de madres diabéticas y sanas. MÉTODOS: Estudio transversal, realizado con 51 parturientes voluntarias, siendo 20 diagnosticadas con diabetes mellitus gestacional y 31 sin cualquier enfermedad asociada. Se recogieron las muestras de suero y de calostro en ayuno en el postparto inmediato y se analizó el α-tocoferol por cromatografía líquida de alta eficiencia (CLAE). Para definir el estado nutricional de vitamina E, se adoptó el punto de corte sérico (697,7µg/dL). El análisis estadístico se realizó con la prueba t de Student para variables independientes a fin de comparar las concentraciones medianas de α-tocoferol en el suero entre los grupos control y con diabetes mellitus gestacional. Ese análisis también fue realizado para comparar los promedios de α-tocoferol en el calostro en los grupos estudiados. Se utilizó la correlación de Pearson para evaluar la relación entre la concentración de α-tocoferol en el suero y en el calostro para ambos grupos. Las diferencias fueron consideradas significativas cuando p<0,05. RESULTADOS: Las concentraciones de α-tocoferol en el calostro fueron 1.483,1±533,8µg/dL para las mujeres del Grupo Control y 1.368,8±681,8µg/dL para las diabéticas, no habiendo diferencias (p=0,50). Sin embargo, en el suero de las puérperas control, la concentración de α-tocoferol fue 1.059,5±72,7µg/dL y, en las diabéticas, 1.391,4±531,5µg/dL, con p<0,01).
